Intervention
UBX0101
Mechanism, regulatory status, and an honest, tiered evidence map.
▲ Human lifespan evidence: no lifespan RCT — a phase 2 human trial (knee osteoarthritis) failed its primary endpoint
What it is
Class: p53/MDM2-interaction inhibitor (local senolytic)
Also known as: UBX-0101
Relationship to senescence: A small molecule that disrupts the p53/MDM2 interaction to push senescent cells into apoptosis; developed for local intra-articular delivery in osteoarthritis.
Regulatory status
Investigational; not approved. Development as a knee-osteoarthritis senolytic was discontinued after a failed Phase 2 trial.
Mechanism
Intra-articular injection intended to clear senescent cells in the joint. See /p53-mdm2.
Evidence — Human (Phase 2, randomized)
| Species / population | Adults with painful moderate-to-severe knee osteoarthritis (n=183). |
| Exposure, route, schedule | Single intra-articular injection of UBX0101 (dose-ranging) vs placebo. |
| Comparator / duration | Randomized, double-blind, placebo-controlled. |
| Endpoint / numeric result | Change in WOMAC-A pain at 12 weeks — no statistically significant difference vs placebo in any arm. |
| What it did NOT establish | No benefit demonstrated; program discontinued. Company cited an unusually strong placebo response. |
Negative or null findings
- Primary endpoint (WOMAC-A at 12 weeks) not met; UNITY Biotechnology did not advance UBX0101 to pivotal studies.
